4basebio is an innovative gene synthesis company with the ambition to provide high quality, synthetic DNA for use in gene therapy and gene editing platforms. The company is growing rapidly and is establishing a GMP gene synthesis hub at its site north of Cambridge, UK. We are looking for dynamic and experienced team members to help us achieve our ambitions and goals.

We are expanding and looking to hear from Scientists which experience in any of the following skill sets:

1) DNA purification techniques
• Precipitation
• Proficient in use of AKTA FPLC systems
• IEX, HIC, Affinity chromatography
• Tangential Flow filtration (TFF)/ cross flow filtration

2) DNA/RNA synthesis/ analysis
• Rolling circle DNA amplification
• qPCR and RT-qPCR
• DNA Ligation
• DNA Restriction enzymes
• Molecular biology and DNA construct design
• Cloning, transformations and E. coli fermentation
• Plasmid maxi preps
• In vitro transcription
• RNA purification
• DNA analysis:
- AGE
- Qubit
- UV/VIS
- Mass Spec
- HPLC
- Blotting

3) Tissue culture/nanoparticles
• Extensive tissue culture experience in relevant biological models
• Nanoparticle formulation/microfluidics
• Nanoparticle characterisation (DLS, microscopy, nano-tracking analysis)
• ELISAs
• Western blotting
• In vivo experience

4) Gene editing
• Experience in gene editing techniques such as CRISPR/Cas9, TALENs
• Construct design of repair templates
• Application of editing strategies in mammalian cells/ primary cells
• Delivery mechanisms- viral non-viral or electroporation  

5) AAV manufacture
• AAV production
• AAV purification
• AAV characterisation – TCID50, full:empty ratio, ELISA, RTqPCR

You will hold preferable a First Degree in Life Sciences or Biotechnology
