PHAGE LIBRARY SCIENTIST (2-Year Fixed Term Contract)

Bicycle Therapeutics (NASDAQ: BCYC) is a clinical-stage biopharmaceutical company developing a novel class of medicines, referred to as Bicycles®, for diseases that are underserved by existing therapeutics. Bicycles are fully synthetic short peptides constrained with small molecule scaffolds to form two loops that stabilize their structural geometry. This constraint stabilizes the structural geometry of the peptide and facilitates target binding with high affinity and selectivity, making Bicycles attractive candidates for drug development. Bicycle’s lead product candidate, BT1718, is a Bicycle Toxin Conjugate being investigated in an ongoing Phase I/IIa clinical trial in collaboration with the Centre for Drug Development of Cancer Research UK. Bicycle is headquartered in Cambridge, U.K. and has an office in Lexington, MA. As our team in the UK expands, we have created a new opportunity in our Discovery Biology team. The successful candidate will join a dynamic laboratory team to further develop Bicycle’s core technology. The role involves library design, generation and testing a as well as driving and innovating across Bicycle’s phage platform.

We are looking for a highly motivated, methodical, well organised Research Associate or Research Scientist who enjoys working at the bench and performs consistently at the highest level. Previous molecular biology experience is essential; experience of working with phage highly desirable, but not essential as training can be provided.

• Molecular biology to prepare key complex reagents

o Large scale preparation of Bicycle’s unique phage libraries

o Preparation of plasmid DNA, restriction cloning

o Electroporation/chemical transformation of E. coli

o Preparation of other complex reagents and buffers

• Assay design and implementation to quality control samples

o SDS-PAGE, Western blot, agarose DNA gel analysis

o Quantification of samples by qPCR, UV absorbance

o Biochemical assays e.g. AlphaScreen, ELISA

• Innovation and development of core platform technology

o Conceive, design and reduce to practice innovations in the phage platform technology to drive library generation efficiency and improve library diversity

o Conceive, design and reduce to practice improvements to phage panning and selection of Bicycle peptides

• Data Integrity and communication

o Accurate record keeping via electronic lab notebook system

o Presentations to management and colleagues

Essential Skills and Competencies:

• A BSc in a biological or chemical subject with experience of working in a pharmaceutical/biotech laboratory setting

• Significant demonstrable molecular biology laboratory experience

o Cloning, PCR

• Ability to plan, conduct and analyse experimental work independently

• Strong interpersonal skills; effective communication with multidisciplinary teams both locally and remotely is essential

• Proactive approach to problem solving and strong self-motivation

• Excellent organisational skills and close attention to detail

• Diligence in documenting work and understanding of the importance of data integrity

• Proficiency with MS Office

Desirable Capabilities:

• Working knowledge of phage display or other display technology

• Experience of biochemical in vitro assays e.g. enzyme inhibition assay or ELISA

• Experience of working in matrix environment as part of cross-functional team

• Experience of using electronic lab notebook systems e.g. Dotmatics
