Spun out from the Karolinska Institute in 2013, Pelago Bioscience AB was founded by the inventors of the Cellular Thermal Shift Assay (CETSA®) to both develop the method and to provide a commercial service for organisations wanting to use the method.. . CETSA measures compound binding under actual physiological conditions within either a cellular of lysate environment. The assay relies on a well-established observation that compound binding with a protein will change its thermal denaturation point, meaning that at a given denaturation temperature there will be a difference in the abundance of the intact protein as compared with a negative control. It is the only technique that will confirm such target engagement is occurring and provides ‘real world’ EC50 values for your compound in cells.. .
